In Vitro Effect of Erythropoietin on the Spleen of The Polycythemic Mouse
نویسنده
چکیده
I N VIEW OF THE FUNCTION of hemopoietic stem cells maintaining a hematologically steady state, radiosensitivity of the stem cells has been regarded as responsible for postirradiation hemopoietic death. As a method for studying the effect of irradiation upon hematopoietic stem cells, the colonyforming cells first described by Till and McCulloch’ 3 and the erythropoietin responsive cells in polycythemic animals4’5 have been used. The present study was performed for the purpose of developing an in vitro method to observe the radiosensitivity of stem cells. The previously reported tissue culture method of estimating the response of polycythemic mouse spleen to erythropoietin6 was used to observe an in vitro and in vivo radiosensitivity of hematopoietic cells and the recovery from irradiation.

Transplantation and Homing of Mouse Embryonic Stem Cells Treated with Erythropoietin in Spleen and Liver of irradiated mice
Background: The present study was designed to evaluate the homing potential of mouse embryonic stem cells (ESC) treated with erythropoietin (EPO) in hematopoietic organs such as spleen and liver after transplantation using morphological and immuno-histochemical techniques. Methods: Day-four embryoid body (EB)-derived cells were dissociated and re-plated in medium in the presence and absence of ...
متن کاملDifferentiation of Mouse Yolk Sac Cells to Erythroid Cells in The Presence of Erythropoietin
Purpose: Yolk sac hematopoietic stem cells (YS-HSC) have two dominant characteristices: a larger reproductive capacity and the absence of the expression of MHC associated antigens. Therefore, these cells are promising candidates for transplantation, cell therapy and gene manipulation. There are controversial reports on the effects of erythropoietin (EPO) on the differentiation of yolk sac cells...
متن کاملEffect of serum starvation stress on the mouse spleen mononuclear cells mixed culture: Introducing a new immunomodulatory method
Introduction: Bone marrow and immune cell transplants are a common method of treating some blood diseases around the world. However, due to the malfunction of the immune system, its use is not always effective. In this study, we evaluated the potential of culture stress in serum-free medium in regulating the immune system. Materials and Methods: Spleen immune cells were isolated from Balb/C and...
متن کاملEffect of testosterone on the formation of erythroid spleen colonies from fetal liver precursor cells.
Polycythemic x-irradiated female mice, injected with 12-day fetal liver cells, showed 81#{176}/o suppression of erythroid spleen colonies as compared with nonpolycythemic recipients. On the other hand, in male recipients only 16#{176}/osuppression was observed. Hence, androgenic hormones seem to play a role in regulating erythropoiesis of explanted fetal stem cells. To test this, we examined th...
